Can I use a partitioned USB drive as extended storage?

I already have a backup USB 3.0 drive that I use for PC backup purposes. Can I create a 250+ GB partition on the drive and format that partition on my PS4 to use as extended storage, or will the PS4 reformat the whole drive?
